IgG extracted from dog serum is mixed with ammonium sulphate in a tube and centrifuged to obtain the precipitate which is diluted with PBS and dialysed overnight and again centrifuged to get the supernatant which is considered as the Lowry sample and then protein concentration is calculated from the Lowry Method. On the next day, Column Chromatography is done after washing thoroughly and readings of the samples have been taken under UV-Spectrophotometer. After that SDS-PAGE has been done and bands are obtained in the gel and then (absorbance vs logMW) graphs of the respective bands of SDS-PAGE have been done.
